In order to evaluate the potential use of environmental scanning electron microscopy (ESEM) in biology, structural changes of the jejunal villi of rats were studied after periods of fasting and refeeding, using a conventional scanning electron microscope (CSEM) and ESEM. While observation using the CSEM, involves chemical fixation, drying and coating, observation of fresh, unprepared materials can be directly realized with the ESEM. Environmental microscopy provides a relatively new technology for imaging hydrated materials without specimen preparation and conductive coating. Direct observation of biological samples in their native state is therefore possible with an ESEM.

After fasting, the jejunal mucosa is dramatically reduced in size, splits and holes appearing at the tip of the villi. These changes were observed whatever the type of technique used. Artifacts due to the sample preparation for CSEM observation (drying, coating) can therefore be excluded. However, CSEM and ESEM must be used jointly. While, CSEM must be preferred for surface analysis involving high magnifications, ESEM observation, on the other hand, can prove valuable for determining the living aspect of the samples.  相似文献

Relying on first principles simulations of stoichiometric MgO, ZnO, and NaCl (1x1) ultrathin (111) films, we demonstrate the existence of a critical thickness below which polarity is uncompensated: the surface charges are bulklike, and the total dipole moment and the formation energy grow linearly with thickness. This study reveals novel facets of the problematics of polarity akin to the nanoscopic size of the objects and opens stimulating perspectives on polar nanostructures with surface properties and reactivity unaffected by charge compensation as in macroscopic samples.  相似文献

The antitumour activity of the organometallic ruthenium(ii)-arene mixed phosphine complexes, [Ru(eta(6)-p-cymene)Cl(PTA)(PPh(3))]BF(4) and [Ru(eta(6)-C(6)H(5)CH(2)CH(2)OH)Cl(PTA)(PPh(3))]BF(4) (PTA = 1,3,5-triaza-7-phosphaadamantane), have been evaluated in vitro and compared to their RAPTA analogues, [Ru(eta(6)-p-cymene)Cl(2)(PTA)] and [Ru(eta(6)-C(6)H(5)CH(2)CH(2)OH)Cl(2)(PTA)] . The results show that the addition of the PPh(3) ligand to increases the cytotoxicity towards the TS/A adenocarcinoma cancer cells, which correlates with increased uptake, but also increases cytotoxicity to non-tumourigenic HBL-100 cells, thus decreasing selectivity. The decrease in selectivity has been correlated to increased DNA interactions relative to proteins, demonstrated by reactivity of the compounds with a 14-mer oligonucleotide and the model proteins ubiquitin and cytochrome-c.  相似文献

This work deals with the characterization of the kinematic work-hardening of a bake-hardening steel. A shear test device has been designed and its use for the characterization of the work-hardening of sheet metals is described. Two main results are presented. Firstly, a local strain measurement, based on the following of three dots drawn on the gauge area, gives the evolution of the strain tensor eigenvalues during the test. It is shown, by comparing the theoretical kinematics of simple shear with a slightly perturbated one, that the strain state is close to the ideal one in the center of the gauge area. Secondly, reversal of the shear direction is performed after several prestrain and the evolution of the kinematic work-hardening with the equivalent plastic strain has been identified using an anisotropic elasto-viscoplastic model of Hill 1948 type. Isotropic and kinematic contributions of the work-hardening are also calculated from loading–unloading tensile tests and are compared to those obtained from the simple shear tests. The results show a discrepancy between both identification for the isotropic and the kinematic hardening. However, they are in agreement concerning the evolution of the global work-hardening.  相似文献

While consequences of frustration of magnetic interactions are much studied in localized spin systems, much less studies have been performed on frustrated metallic systems. However, several effects of strong geometrical frustration in metallic correlated system have also been experimentally observed in rare-earth or transition metal compounds: coexistence of magnetic and non-magnetic sites in ordered magnetic structure, heavy fermion behaviour and anomalous Hall effect due to spin chirality are consequences of frustration. An overview of the experimental observations and of the proposed models is given. Other interesting effects due to magnetic frustration in metallic systems, which have been predicted theoretically, are also reviewed.  相似文献

OBJECTIVE: An exploratory analysis of co-aggregation of cancers using registry-based data. METHODS: We utilized sibships from over 18,000 families who had been recruited to the NCI-sponsored multi-institutional Cancer Genetics Network. The analysis assesses co-aggregation at the individual and family level and adjusts for ascertainment. RESULTS: We found statistically significant familial co-aggregation of lung cancer with pancreatic (adjusted p < 0.001), prostate (adjusted p < 0.003), and colorectal cancers (adjusted p = 0.004). In addition, we found significant familial co-aggregation of pancreatic and colorectal cancers (adjusted p = 0.018), and co-aggregation of hematopoietic and (non-ovarian) gynecologic cancers (adjusted p = 0.01). CONCLUSION: This analysis identified familial aggregation of cancers for which a genetic component has yet to be established.  相似文献
